Napoleon and Snowball (Animal Farm) Essay

George Orwell’s Animal Farm is an allegorical novel that reflects events such as the Bolshevik revolution, the economic reform by Trotsky the Russian famine in 1921 and during the Stalin era before the Second World War. The pigs represent the communist leadership, and the rest of the animals on the farm symbolize the different parts of Russian society and how the communist takeover of Russia affected them. Two main characters, Snowball and Napoleon (who symbolize Trotsky and Stalin), engages in a political struggle, as both of them wanted to have the power to lead all of the animals. Although Napoleon wins the struggle in the end, Snowball’s political strategy is better than the corrupted leadership of Napoleon. Snowball cares more about the living condition of the animals. Compared to Napoleon, Snowball knows the meaning of â€Å"Animalism†, which is animals are equal and it should lead the animals to a better life. Snowball is interested in reading all kinds of books to enrich his scope of knowledge to make the animals work easier. For example, he declares the windmill idea from a book of the Jones. Also Snowball is very brave; he has studied the book of Julian Caesar and successfully led through the Battle of the Cowshed while Napoleon was hiding himself in the barn. Snowball has an intelligent and friendly character. Unlike Napoleon, Snowball never forces someone to agree with his ideas. Instead, he is good at explaining how his ideas work and persuades all the animals to believe and trust his ideas. Unlike Napoleon, Snowball does not need Squealer’s help, he can do the explaining part all by himself. Napoleon has his own ways to control other animals such as the dogs and Squealer but Snowball has different way which is the committees. Snowball seems to work better within the political system. Napoleon goes around it. Napoleon, for instance, he understands the role of force in political control and uses his attack dogs to expel Snowball from the farm. Napoleon seems to have a powerful, self-centered desire for control, while Snowball seems to think of himself as a genius who should be the one to guide the farm toward success. Snowball is the only one who knows the real meaning of â€Å"Animalism†, and he is putting out true effort to make the living standard of the animals better than before, and insists everyone is equal. After the expel of Mr. Jones, the Animal Farm is supposed to be democracy, all of the animals should decide how to do things together, any one animal to rise to greater power than any other would violate that ideal, and make Animal Farm indistinguishable from a human farm. On the other hand the only thing that Napoleon thinks about is how to gain more power and personal benefit from the farm. Snowball always comes up with some long-term plans and on the other hand Napoleon can only come up with some short-term plans. In conclusion, Snowball’s political strategy is better then the corrupted leadership of Napoleon.

Feminism And Gender Equality In The 1990’s

Overall, the rights and status of women have improved considerably in the last century; however, gender equality has recently been threatened within the last decade. Blatantly sexist laws and practices are slowly being eliminated while social perceptions of â€Å"women's roles† continue to stagnate and even degrade back to traditional ideals. It is these social perceptions that challenge the evolution of women as equal on all levels. In this study, I will argue that subtle and blatant sexism continues to exist throughout educational, economic, Women who carefully follow their expected roles may never ecognize sexism as an oppressive force in their life. I find many parallels between women's experiences in the nineties with Betty Friedan's, in her essay: The Way We Were – 1949. She dealt with a society that expected women to fulfill certain roles. Those roles completely disregarded the needs of educated and motivated business women and scientific women. Actually, the subtle message that society gave was that the educated woman was I remember in particular the searing effect on me, who once intended to be a psychologist, of a story in McCall's in December 1949 called â€Å"A Weekend with Daddy. A little girl who lives a lonely life with her mother, divorced, an intellectual know-it-all psychologist, goes to the country to spend a weekend with her father and his new wife, who is wholesome, happy, and a good cook and gardener. And there is love and laughter and growing flowers and hot clams and a gourmet cheese omelet and square dancing, and she doesn't want to go home. But, pitying her poor mother typing away all by herself in the lonesome apartment, she keeps her guilty secret that from now on she will be living for the moments when she can escape to that dream home n the country where they know â€Å"what life is all about. † (See I have often consulted my grandparents about their experiences, and I find their historical perspective enlightening. My grandmother was pregnant with her third child in 1949. Her work experience included: interior design and modeling women's clothes for the Sears catalog. I asked her to read the Friedan essay and let me know if she felt as moved as I was, and to share with me her experiences of sexism. Her immediate reaction was to point out that â€Å"Betty Friedan was a college educated woman and she had certain goals that never interested me. My grandmother, though growing up during a time when women had few social rights, said she didn't experience oppressive sexism in her life. However, when she describes her life accomplishments, I feel she has spent most of her life fulfilling the expected roles of women instead of pursuing goals that were mostly reserved for men. Unknowingly, her life was controlled by traditional, sexist values prevalent in her time and still prevalent in the nineties. Twenty-four years after the above article from McCall's magazine was written, the Supreme Court decided whether women should have a right to an abortion in Roe v. Wade (410 U. S. 113 (1973)). I believe the decision was made in favor of women's rights mostly because the court made a progressive decision to consider the woman as a human who may be motivated by other things in life than just being a mother. Justice Blackmun delivered the Maternity, or additional offspring, may force upon the woman a distressful life and future. Psychological harm may be imminent. Mental and physical health may be taxed by child care. There is also a distress, for all concerned, associated with the unwanted child, and there is the problem of bringing a child into a family lready unable, psychologically and otherwise, to care for it. In other cases, as in this one, the additional difficulties and continuing stigma of unwed motherhood may be involved. I feel the court decision of Roe v. Wade would not have been made in 1949. Even in 1973, it was a progressive decision. The problem of abortion has existed for the entire history of this country (and beyond), but had never been addressed because discussing these issues was not socially acceptable. A culture of not discussing issues that have a profound impact on women is a culture that encourages women to be powerless. The right of abortion became a major issue. Before 1970, about a million abortions were done every year, of which only about ten thousand were legal. Perhaps a third of the women having illegal abortions – mostly poor people – had to be hospitalized for complications. How many thousands died as a result of these illegal abortions no one really knows. But the illegalization of abortion clearly worked against the poor, for the rich could manage either to have their baby or to have their abortion under A critic of the women's movement would quickly remind us that omen have a right to decline marriage and sex, and pursue their individual interests. However, I would argue that the social pressure women must endure if they do not conform to their expected role is unfair. The problem goes beyond social conformity and crosses into government intervention (or lack thereof). The 1980's saw the pendulum swing against the women's movement. Violent acts against women who sought abortions became common and the government was unsympathetic to the victims. There are parallels between the Southern Black's civil rights movement and the women's movement: Blacks have long been ccustomed to the white government being unsympathetic to violent acts against them. During the civil rights movement, legal action seemed only to come when a white civil rights activist was killed. Women are facing similar disregard presently, and their movement is truly one for civil rights. A national campaign by the National Organization of Women began on 2 March 1984, demanding that the US Justice Department investigate anti-abortion terrorism. On 1 August federal authorities finally agreed to begin to monitor the violence. However, Federal Bureau of Investigation director, William Webster, declared that he saw no evidence of â€Å"terrorism. † Only on 3 January 1985, in a pro-forma statement, did the President criticize the series of bombings as â€Å"violent anarchist acts† but he still refused to term them â€Å"terrorism. Reagan deferred to Moral Majoritarian Jerry Falwell's subsequent campaign to have fifteen million Americans wear â€Å"armbands† on 22 January 1985, â€Å"one for every legal abortion† since 1973. Falwell's anti- abortion outburst epitomized Reaganism's orientation: â€Å"We can no longer passively and quietly wait for the Supreme Court to change their mind or for Congress to pass a law. Extremism on the right was no vice, moderation no virtue. Or, as Hitler explained in Mein Kamph, â€Å"The very first essential for success is a perpetually constant and regular employment of violence. (See This mentality continued on through 1989 during the Webster v. Reproductive Health Services (109 S. Ct. 3040 (1989)) case. â€Å"The Reagan Administration had urged the Supreme Court to use this case as the basis for overturning Roe v. Wade. † (See Endnote #5) It is disturbing that the slow gains achieved by the women's movement are so volatile and endangered when conservative dministrations gain a majority in government. To put the problem into perspective: a woman's right to have an abortion in this country did not come until 1973. Less than two decades later, the president of the United States is pushing to take that right away. It seems blatant that society is bent on putting women in From the above examples, it appears American culture prefers women as non- professional, non-intellectual, homemakers and mothers. This mentality is not easily resolved, because it is introduced at a young age. Alice Brooks experienced inequality on the basis of her race and her sex. In her autobiography, A Dream Deferred, she recalls the reaction of her father when she brought up the idea of college to him: I found a scholarship for veterans' children and asked my father to sign and furnish proof that he was a veteran. He refused and told me that I was only going to get married and have babies. I needed to stay home and help my mother with her kids. My brother needed college to support a family. Not only was I not going to get any help, I was also tagged as selfish because I wanted to go This is another example of women being labeled as selfish for wanting the same opportunities as men. Alice Brooks is a very courageous woman; seemingly able to overcome any oppression she may encounter. During her presentation to our class, she said that â€Å"women who succeed in male dominated fields are never mediocre – they are extraordinary achievers. Her insight encapsulates much of the subtle sexism that exists today. I feel that no one can truly be equal in a society when only the â€Å"extraordinary achievers† are allowed to succeed out of their This attitude of rising blatant and subtle attacks on women's civil rights is further exemplified in recent reactions to affirmative action plans. These plans have been devised to try to give women and minorities an opportunity to participate in traditionally white male dominated areas. However, we see the same trends in legal action for the use of affirmative action plans as we saw in the 1980's backlash against the Roe v. Wade decision. A few interesting points were presented in the case, Johnson v. Transportation Agency, Santa Clara (480 U. S. 616 (1987)). Mr. Paul E. Johnson filed suit against the Santa Clara County Transportation Agency when he was denied a promotion, feeling the company's affirmative action plan denied him of his ivil rights. Some interesting facts were presented in this case: Specifically, 9 of the 10 Para-Professionals and 110 of the 145 Office and Clerical Workers were women. By contrast, women were only 2 of the 28 Officials and Administrators, 5 of the 58 Professionals, 12 of the 124 Technicians, none of the Skilled Crafts Workers, and 1 – who was Joyce – of the 110 Road Maintenance Workers. (See Endnote # 7) The above statistics show women have been considerably underrepresented at the Santa Clara County Transportation Agency. These numbers are not uncommon and are found throughout business. It is interesting to note the current popular perception is that affirmative action precludes white males from finding employment with companies that implement these plans. The truth is in the numbers, however. The fact that Mr. Johnson felt he was denied his civil rights because an equally qualified woman was given a promotion, instead of him, is just a small window into the subtle sexism that exists today. Most critics of affirmative action do not consider the grossly unequal numbers of men in management and professional positions. Secondly, it never seems an issue of debate that a woman may have had no other previous life pportunities in these male dominated areas. I do not intend to argue that affirmative action is good or bad, but only wish to point out that the current backlash against these programs is heavily rooted in sexism and racism. Often blatant violence or unfair acts against a group of people will cause that group to pull together and empower themselves against their oppressors. The women's movement has made large steps to eliminate many of these blatantly sexist acts in the last century. Now the real difficulty is upon us: subtle acts of sexism and the degrading social roles of women in today's onservative culture. Alice Brooks so eloquently described her experiences with inequality, stating, â€Å"the worse pain came from those little things people said or did to me. † As these â€Å"little things† accumulate in the experience of a young woman, she increasingly finds herself powerless in her relationships, employment, economics, and society in general. The female child has as many goals as the male child, but statistically she is unable to realize these goals because of the obstacles that society sets in front of her. Society and media attempt to create an illusion that women have every right that men enjoy.

A problem occurs as those sugars might transform to an alcohol named ethanol (or ethyl alcohol) by natural fermentation process which is not performed by enzymes. According to scientific review, both natural and manufactured products contain small amount of alcohol; for example, fruits, juices, vegetables, breads, cheeses, beef, and honey. Those food and beverage usually contain not more than 0. 5% of alcohol. Therefore, anything containing sugar is fermentable into alcohol. Other manufactured products such as Coca Cola, Pepsi, and Mirinda contain alcohol at percentage range of 0. 2% – 0. 3% as Beta Carotene (the colouring used) is melted by using the alcohol method. In addition, according to Eastern Standard Time on July 8, 1999 (4:00 pm); â€Å"The oils that they use to make Pepsi have minute trace of alcohol which combined make up a percentage of alcohol. † The problem of alcohol that might contain in food or beverage has been debated by Mujamma’ Al-Fiqhi Al-Islami as certain types of alcohol are beneficial in food production. According to the Islam guidelines, Muslims are allow consuming ethanol as it is not harmful but only can be taken at small amount which is not more than the specified percentage.

Psychoanalytic Perspectives of the Oedipus Mythology Essay Patricide and incest form the thesis and message that Sophocles began with the creation of Oedipus the King. In the plays that followed, Oedipus at Colonus and Antigone, Sophocles expounded upon what would become one of the most profound archetypes in psychoanalysis. To best interpret the Oedipus trilogy, a look will be taken into the construction of the plays themselves, followed by an interpretation of the plays’ parallels and the inception of the Oedipus Complex based upon a psychoanalytic perspective. To begin with, Sophocles wrote what became known as the Oedipus trilogy over a period of more than forty years which indicates the profound immersion that he plainly had in the Oedipus saga. Each play is a self-contained chronicle representing his dramatic theme of redemption from the sin of patricide and incest, and yet, the arch between the three Theban plays highlights the message that Sophocles refused to relieve himself from, and which consumed nearly his entire life. While this may seem of little importance to reading the Oedipus trilogy itself, Sophocles did not write them in the order represented in nearly every anthology. As David Grene notes, â€Å"as far as the legend is concerned, the story runs in sequence: Oedipus the King, Oedipus at Colonus, Antigone† (p. i). Their order of conception, instead, went: Antigone, Oedipus the King, and then Oedipus at Colonus (p. i), which presents a few inaccuracies within the story itself (mainly with the character and actions of Creon, Jocasta’s brother). From this birth order, â€Å"the series, therefore, cannot have formed a [true or literal] trilogy†¦beyond the fact that each of the three plays deals with the situation in the Oedipodean family history, there is no unity of theme or treatment between them† (Watling). Moreover, â€Å"except for the obvious links of fact connecting them, each constitutes a fresh approach to a distinct and self-contained problem† (13). Roughly, this means that while most anthologies present the three Theban plays in a chronological order for the character Oedipus, the fact remains that each could be read without knowledge of the others and the same theme and message would be received—which, based upon Sophocles’ life-long obsession with the story, must have been his justification for the story that kept evolving. The reasoning behind the order and placement of the plays within anthologies is sound, however, because, while the stories may be self-contained, the arch of Oedipus is the link that literally turns the plays into a trilogy. In Oedipus the King, Oedipus, as prophecy had predicted, kills his father, King Laius, and marries his mother, Queen Jocasta, bearing at least four children in the process who, in the play Antigone, are revealed as Antigone, Ismene, Eteocles, and Polyneices. After learning of his horrible actions, Oedipus exiles himself as he had proclaimed would be the fate of the brute capable of murdering King Laius, and subsequently blinds himself in the hopes of finding redemption for his unforgivable sins. While Oedipus at Colonus is the second play in the anthologies, it is the literal ending for Oedipus. He is taken to the city of Colonus by his loving daughters, Antigone and Ismene, because, as he had learned from the oracle in Oedipus the King, he was meant to find his final resting place there. Moments before his end, Oedipus realizes that his sins of patricide and incest weren’t truly sinful because he committed the acts out of ignorance alone, and it is in this moment, and despite everything he has faced in his quest for redemption that Zeus absolves Oedipus of his sins and he passes, with peace, into the afterlife. In Antigone, Oedipus is little more than a plot reference to get the play started. His only mention is in the opening lines and is that, following his self-imposed exile from the city of Thebes in Oedipus the King, Oedipus had made a prediction that his sons would be arrogant enough to fight over his throne and kill each other. Thus the play begins with the deaths of Eteocles and Polyneices. The play itself follows the actions of Antigone and her husband Creon, who is the cause for much speculation from most scholars due to his not-so advanced age. As the timeline goes, Creon is Jocasta’s brother and would have been Oedipus’ senior by many years or, at the very least, would have been as old as Oedipus himself. As Antigone is Oedipus’ daughter, and Creon is said to be â€Å"a vigorous middle-aged father of a youthful son† (Watling, 13), scholars debate as to the accuracy of Sophocles’ character creation and question as to his motives in retelling the story in as many forms as he did, with an outstanding flaw. Creon’s deviation notwithstanding, it is the minor and insignificant role of Oedipus in the play Antigone that marks the main reason that most anthologies place Antigone last in the order of the Oedipus mythology, even though the timeline would, as Sophocles thought in writing them, place Oedipus at Colonus last, due to the play’s depiction of the last days of Oedipus’ life. This, in itself, is significant in understanding the psychological aspects of the Oedipus trilogy. Now, while readers will never know the true meaning behind the order of the Oedipus mythology, the fact remains that Sophocles wrote within an archetype and character arch with the intentions of presenting his dramatic theme—and it took three tries to get it out as he desired. What he created, however, demonstrates a truth about the self-discovery of the individual and the path to redemption that marks a significant aspect of the growth of the human being. To best define this journey, a psychoanalytic perspective will now be taken into Sophocles’ main theme and message. From an analysis of his work, Sophocles â€Å"shares [a] concern with finding truth in a world of appearances and is influenced, even if indirectly, by the new theories about language: the problem of the relation of words to reality, and the power of words to deceive, to win unjust causes, and to confuse moral issues† (Segal, 7). More, the plays are â€Å"almost certainly a response to events of [his] period. An unexpected, supernatural-seeming disaster suddenly sweeps away brilliant hopes; confidence in human reason and calculation is shattered, and greatness swiftly turns into misery† (9). Sophocles saw the rise and fall of powerful nations, and it makes sense that he would take what he had seen and created his rendition of events that had transpired in a literary form that he could show the world. And it is from this basis that scholars have debated over the purpose and parallels behind the two main cities present within the plays of Sophocles. Often, the city of Athens is compared to a man’s relationship with himself, whereas the city of Thebes represents the conflict between man and his father. Indeed, â€Å"the figure of Oedipus [is] a distillation of Athens at the height of its power, energy, daring, intellectual curiosity, and confidence in human reason† (Segal, 11). As the historical context of the city of Athens was surely an influence in Sophocles’ making of the Oedipus trilogy, a direct parallel from the destruction of Athens to the destruction of Oedipus the powerful leader can be drawn. More, â€Å"it is even possible that Oedipus’ search for who he really is reflects something of a communal identity crisis in a city that had undergone a massive transformation in a short time and had refashioned itself from a rather quiet, traditional aristocracy and tyranny in the sixth century into a radical, intellectualized democracy and a powerful empire† (Segal, 11). This parallel, of Oedipus to the grand city of Athens, does much to lend weight to Sophocles’ theme of destiny and the gods marking a path for man. For, as Athens rose in power, so too, it fell because it committed sins and transgressions to great to find redemption from along the way. As for the city of Thebes, there lies a direct correlation to Sigmund Freud’s theory of conflict between man and his father which represents a direct parallel to Oedipus’ exile from the city of his king-making. Freud suggested that â€Å"the play fascinates us so much†¦not because it dramatizes ‘the contrast between destiny and human will,’ but because ‘there must be something which makes a voice within us ready to recognize the compelling force of destiny’† (Segal, 59). From this theory, Freud defined that the â€Å"‘destiny’ is the universal necessity to which all of us (or at least all males) are subject—namely, the wishes that remain from our buried animal nature to kill the father and possess the mother† (59). And it is this destiny upon which Sophocles created his foundation and archetype. As a basis for his theory, Freud determined that the oracle was a direct parallel to the subconscious mind, citing that â€Å"this disguising of Oedipus’ unconscious desires in the form of an oracle from the gods not only lets the unconscious become visible but also accounts for the feeling of guilt that we have about these unconscious desires, even though we are not guilty of any crime† (Segal 59-60). Even more, Freud speculated that â€Å"Oedipus’ eagerness to punish himself, with no attempt at self-defense, corresponds to the inner conviction of guilt that stems from these unconscious desires† (60). Because Oedipus immediately set out to punish himself and find redemption for his actions, despite the fact that he did them in ignorance, then, suggests that in his subconscious, Oedipus did indeed have desires to kill his father and physically and sexually possess his mother. For, as Freud theorized, in his actions following his revelation, Oedipus confessed his own guilt. If, perhaps, Oedipus had sought to defend himself on the basis that he didn’t know his father and certainly had no affections for his mother, having grown up the adoptive son of another, the theme may then have been interpreted differently. However, as Freud was so certain, there is much to be said about the actions of Oedipus in correlation to the guilt he felt. The subconscious mind had a profound impact on Oedipus, even though he knew nothing of the birth parents he was prophesized to destroy. For Freud, the archetypal constructs of the Oedipus mythology was so absolute that he dubbed it the â€Å"‘Oedipus Complex’†¦[which] denotes each person’s attitudes and behavior in his or her most intimate family relationships, especially to mother and father†¦[and, to mature into a normal adult, the child] must somehow come to terms with the residue of repressed infantile hatred and desire for his or her parents† (Segal, 60). The Oedipus Complex, in modern psychology, is applied to the study of actions in relation to the subconscious desires that exist in every child. Even more, Freud’s theory has become common in the vernacular of psychology to the extent that scholars use the term, perhaps, without even grasping the full meaning behind the cleverly, yet aptly, named psychosis. The Oedipus Complex is based upon two main ideals: that of the matriarchal relationship to the child and the patriarchal relationship. The two are separated by this relationship and define the growth and development of the child. Moreover, â€Å"matriarchal culture is characterized by the emphasis on ties of blood, ties to the soil and the passive acceptance of all natural phenomena†¦[while a] patriarchal society in contrast is characterized by respect for man-made law, by the predominance of rational thought and by the effort to change natural phenomena by man† (Armens, viii). To better define the difference, â€Å"in the matriarchal concept all men are equal since they are all the children of mothers and each one a child of Mother Earth. A mother loves her children all alike and without (limiting) conditions†¦the aim of life is the happiness of man and there is nothing more important or dignified than human existence and life† (viii). In this, to draw a parallel back to Oedipus, the child has (whether he realizes or understands it) a desire to be with the woman who created him because of her earth-mother nature. The desire of every man is to be with a woman who understands everything and will love unconditionally—and that figure, from the start of life, happens to be the mother. However, â€Å"the patriarchal system, on the other hand, recognizes obedience to authority as its main virtue. The principle of equality is replaced by a hierarchical order in society and state, ruled by an authority just as the family is dominated by the father† (Armens, viii). It is because of this very concept that boys contain within them (whether known to the conscious mind or not) the desire to overthrow their father and become the leader of the hierarchy. More, men, by their very nature, contain the desire to become like their fathers, to achieve the power that they may possess, to be the biggest, strongest, fastest titan in the industry. With a full understanding of the Oedipus Complex, a final parallel can be drawn to the path of self-discovery that Oedipus’ entire life is consumed with. As Sophocles had directed, Oedipus â€Å"dramatizes the lonely path of self-discovery† (Segal, 13). And it is in this path that the true nature of Oedipus is revealed. For, the path that he must take is a perilous expedition, not only through the ascent of age, but in the self discovery that every man must achieve to live a fulfilling and successful life. However, it is on this path that Oedipus also meets the very destiny that his parents had hoped to terminate with their preemptive strike. In this, Oedipus is the â€Å"paradoxical combination of knowledge, power, and weakness† (13). He is strong as a king and husband, yet, when he learns the truth of his actions, he crumbles into a despair so deep that it consumes his entire life—and it isn’t a short one. Unequivocally, Sophocles began his thesis with the creation of Oedipus the King and introduced what would become one of the most profound archetypes in psychoanalysis. Throughout the trilogy, the archetypes that Sophocles presents â€Å"[become] a profound meditation on the questions of guilt and responsibility, the order (or disorder) of our world, and the nature of man† (Segal, 12). More, â€Å"the play stands with the Book of Job, Hamlet, and King Lear as one of Western literature’s most searching examinations of the problem of suffering† (12). In looking at the events that transpired throughout the three Theban plays, Sophocles’ main theme that, in any life, destiny and fate will create a man’s destiny; more, the gods have the divine right to tamper with a man’s destiny in the hopes that he will, one day, find redemption from his sins, is rendered by the Oedipus arch throughout the plays. Overall, the three Theban plays, Oedipus the King, Oedipus at Colonus, and Antigone, form the trilogy that founded the most profound archetype in literature and psychoanalysis. Sophocles created the character of Oedipus to highlight his theme of self-discovery and the path to redemption, making it clear that Oedipus, despite his ignorance, would repent until his final moments for his unforgivable sins. However, it is in those final moments that Oedipus finds true absolution and redemption for the subconscious guilt that placed the fantasy of patricide and incest into his desires. And, based upon a psychoanalytic perspective, the theory behind the Oedipus Complex renders the message and main theme of Sophocles’ Oedipus mythology. Works Cited. Armens, Sven. Inhibition Effectiveness of Au Compounds Function of the â€Å"Guiding Bar† on Inhibition Effectiveness of Au Compounds on Thioredoxin Reductase 3 Qi Liu Introduction Mammalian thioredoxin reductase (TR) is an Nicotinamide adenine dinucleotide phosphate (NADPH) dependent flavoprotein oxidoreductase, which is involved in reducing the disulfide of thioredoxin (Trx) using NADPH (Figure 1). Trx can reduce many important proteins, such as ribonucleotide reductase (RNR), which produces deoxyribonucleotides for DNA synthesis1, 2. Thus, TR and Trx play an important role in maintaining proteins in their reduced state, which involves the regulation of cellular redox reactions, proliferation, and differentiation3. In addition, Trx and TR is overexpressed in a lot of aggressive tumors, and the tumor progression and metastasis appear to be dependent on the Trx system, because tumor cells need constant DNA synthesis. However, tumor progression and metastasis were dramatically reduced when TR knockdown cells were injected into mice4. Thus, the TR was proposed to be a new anticancer drug target5. In mammals, there are three different TRs which have been characterized: the cytosolic form TR1, the mitochondrial form TR3, and thioredoxin glutathione reductase (TGR, also known as TR2)6. All of these TRs are selenoproteins with a selenocysteine (Sec) at their C-terminal active site. In the reaction mechanism of TR, the selenolate of Sec acts as an electron donor to Trx, also selenium can accept electrons from the N-terminal redox center of TR6. This redox center is located on a flexible arm, which is solvent-exposed and reactive towards electrophilic inhibitors, thus representing a target for antitumor drug development1. A number of recent reports suggest that gold-based drugs have strong inhibition ability on TRs, which could be from the direct coordination between Au and the active site Sec on TRs, so that the selenolate group is blocked7. Interestingly, the gold compounds,Bis[1,2-bis(dipyridylphosphino)ethane]gold(I) chloride ([Au(d2pype)2]Cl) and Bis[1,3-bis(di-2-pyridylphosphino)propane]gold(I) chloride ([Au(d2pypp)2]Cl), effect the inhibition effectiveness on human TR1(hTR1) and human TR3 (hTR3) differently (Figure2). The precise molecular mechanism of TR inhibition by gold compounds has not been elucidated. Thus, studies on inhibition of TRs by gold-based drugs are necessary for designing new cancer inhibitors. Available crystal structures of TRs include hTR1, rat TR1 (rTR1), mouse TR3(mTR3), DmTR (thioredoxin reductase from D. melanogaster), and PfTR (thioredoxin reductase from P. falciparum) enzymes9. A recent study showed the crystal structure of hTR1 with its substrate thioredoxin, in which the C-terminal tail of hTR1 was stabilized by a â€Å"guiding bar†, so that the flexible C-terminal tail can be observed (Figure 3). The guiding bar was first proposed in one of the structure studies of hTR1, and is composed of three amino acids: Trp407, Asn418, and Asn419. It functions to suppress random motions and positions the C-terminal tail in catalytically competent position through hydrogen bonding interactions9. However, the crystal structure of mTR3 does not show the C-terminus because of the random motion of the C-terminal without control from the guiding bar. Furthermore, the sequence comparison between hTR1 and hTR3 shows the absence of the guiding bar in TR3 (Figure 4). Research question Though inhibition of gold compounds on TRs has been studied, the function of the guiding bar on the gold compounds inhibition effectiveness on TRs has never been brought up to the studies of inhibition effectiveness on TRs. The goal of this proposal is to investigate the role of the guiding bar on the inhibition ability of gold based compounds on hTR1 and hTR3. The guiding bar will be constructed on hTR3 by mutations of K432W, A443N and S444N. Biophysical characterization will be applied to observe the presence of the C-terminus on TR3 after mutagenesis. My hypothesis is the C-terminal tail of the crystal structure of mutated TR3 will be observed, because the random motion of the C-terminus is reduced by restriction from the guiding bar. Then, two gold compounds, [Au(d2pype)2]Cl and [Au(d2pypp)2]Cl , will be applied to test the inhibition difference on normal TR3 and mutated TR3. I expect to see that the inhibition on mutated TR3 could be stronger than that on normal TR3. Significance of Proposed Research The thioredoxin system plays an important role in the intracellular redox enviornment and is composed of Trx and TR10. Proposed studies will give an insight into the function of the guiding bar on different TR enzymes. The guiding bar is expected to suppress the random motion of the C-terminal tail so that the redox center Sec on the C-terminus can have better coordination with the gold compounds. The coordination between metal and Sec will tightly block the Sec redox center, so the intracellular redox balance will be disturbed and result in potent TR inhibition4. Thus, the presence of the guiding bar can help TR to be a better drug target. Proposed studies Enzyme Expression and Purification Recombinant human TR1 and TR3 will be cloned into E. coli BL21(DE3) cells, and the growth and purification will be done as described previously.11 Mutation of Human TR3 In order to study the impact of the guiding bar residues on TR catalysis, the proposed mutations will be constructed on hTR3 with K432W, A443N, and S444N. These three mutation sites are chosen based on the sequence alignment of hTR1 in the guiding bar region of Pro376-Tyr422 (Figure 3), in which the critical amino acids, Trp407, Asn418 and Asn419, have the function of limiting random motion of the C-terminus. Thus, the same function of the guiding bar on hTR3 is expected to be observed via specific site mutation. Mutations will be introduced by the Quick Change mutagenesis method and confirmed by DNA sequencing. Structural Studies of Mutated Human TR3 The C-terminal tail in human and rat TR1 could be observed by X-ray crystallography, because the guiding bar limits its random motion. However, because the guiding bar is not present in mouse TR3, the C-terminal tail is too mobile to generate sufficient electron density without restriction from the guiding bar. This leads to the absence of the C-terminal tail in the crystal structure of mouse TR39. The mutated hTR3 after insertion of the guiding bar will be studied by X-ray crystallography. The presence of an ordered C-terminal tail in the crystal structure and the interaction between the guiding bar and the C-terminal tail will suggest restricted motion of the C-terminal tail. I expect to see that the interaction of the guiding bar on the C-terminal tail will improve the inhibition effectiveness of Au compounds on hTR3. Electrospray ionization mass spectroscopy (ESIMS) Characterization ESI is an ionization technique which is used to detect high molecular weight molecules, such as proteins, peptides and other macromolecules, so ESI MS is an important technique for studying a complex biological sample9. Instead of fragmenting the macromolecules into smaller charged particles, this process turns the macromolecules into small droplets by ionization, and these droplets are further desolvated into even smaller droplets, which creates molecules with attached protons12. As a putative target for anticancer metallodrugs, the selenoenzyme TR is the drug target for gold compounds, such as [Au(d2pype)2]Cl and [Au(d2pypp)2]Cl. The TR inhibition is thought to occur through direct binding of the gold to the active site Sec following ligand substitution. So, TR3 and mutated TR3 will be separately incubated with the two gold compounds mentioned above for 30 mins at room temperature, and then the products will be analyzed by EIS-MS. The results will be compared and are expected to explain the binding of the gold onto the hTRs. I hope to see the gold compounds have better binding on mutated hTR3 than normal hTR3. This can be explained as the guiding bar can reduce the motion of flexible C-terminal tail, so that the gold compounds can bind onto the Sec of mutated hTR3 easier than the normal one and cause more inhibition on the mutated hTR313. TR Inhibition Assay The solvent-accessible selenolate group, arising from enzyme reduction, very likely constitutes a high affinity binding site for gold compounds. Tightly blocking the active site Sec through metal coordination should result in potent TR inhibition9. Thus, after ESI-MS characterization, the TR inhibition study will be performed with the two different gold compounds, [Au(d2pype)2]Cl and [Au(d2pypp)2]Cl, on hTR1, hTR3 and mutated hTR3 respectively. In addition to Trx, some low molecular weight disulfide-containing substrates, including 5,5†²-dithiobis-(2-nitrobenzoic acid) (DTNB), lipoic acid, and lipoamidem, can also be reduced by TRs9. Here, DTNB would be chosen as the substrate for TR inhibition assay to keep consistent with the previous studies from other groups9. The NADPH dependent TR catalyzed reduction of DTNB will be monitored and determined by the increase in absorbance at 412nm. Inhibition of hTR1, hTR3 and mutated hTR3 will be done by using different concentrations of go ld compounds ([Au(d2pype)2]Cl and [Au(d2pypp)2]Cl). The inhibition results from different TRs and gold compounds will be compared. Based on the assumption that the guiding bar can suppress the motion of the C-terminal tail, greater inhibition effectiveness on mutated hTR3 than that on normal hTR3 is expected to be seen. This will show that the C-terminus is limited by the guiding bar on mutated hTR3 helps the gold compounds inhibit the active site Sec more. Binding Studies by Isothermal Titration Calorimetry (ITC) ITC is a technique used to determine the thermodynamic parameters of interaction in solution. It can be applied to study the interactions between protein-protein, enzyme-inhibitor, protein-small molecules, protein-DNA, and so on. Thus, the binding between gold compounds and the three TRs (hTR1, hTR3 and mutated hTR3) can be investigated by ITC. Applying this method will tell us how well the inhibitors, the gold compounds, bind to TRs. The binding affinity (Ka) could show how strongly the gold compounds and TRs will bind, and the higher Ka, the stronger the binding. In addition, enthalpy changes (ΔH) can tell the amount of energy released or obtained. Then, the entropy change (ΔS) and Gibbs energy changes (ΔG) can be determined by the following equation (1): ΔG = -RTlnKa = ΔH-TΔS (1) Comparison of the binding parameters between gold compounds ([Au(d2pype)2]Cl and [Au(d2pypp)2]Cl) and TRs (hTR1, hTR3 and the mutated hTR3) will give more information about the function of the guiding bar on the inhibition effectiveness of TRs. The higher Ka of hTR1 compared to normal hTR3 is expected to be seen, because the absence of guiding bar in hTR3 will lead to a flexible C-terminal tail, which will give a lower inhibition. In addition, Ka of the mutated hTR3 is expected to show higher binding affinity than the normal hTR3, because the guiding bar mutated on hTR3 will help reduce the random motion of the C-terminal tail, so that the redox center Sec can be easily inhibited by the gold compounds. Thus, the guiding bar can help improve the inhibition effectiveness of gold compounds on mutated hTR3. Conclusion Through mutation and structural studies, the presence of the C-terminal tail which is restricted by the inserted guiding bar is expected to be seen in the mutated hTR3. The ESI-MS characterization and binding study can give information about if the gold can successfully bind to the specific active site on the mutated hTR3. Finally, a better understanding of the inhibition effectiveness will be tested and compared by the inhibition assay with hTR1, hTR3 and mutated hTR3. Thus, a better understanding of the function of the guiding bar in the TR system will give an insight into the effect of the guiding bar on the drug inhibition effect on TR3 so that it can become a better drug target. Thus Marlow is able to pronounce that Kurtz is a remarkable individual because he has a value, courage, that Marlow and many other individuals do not posses.

History 10H Mr. Diamond Period 3 The Magic of Walt Disney Walt Disney is considered an icon of American pop culture and has made many contributions to the American entertainment industry. A self-made-man from the Midwest, he became an inspiration to all American children and adults. Hailing from the heart of America, he was very patriotic and contributed a great deal to our country in times of need. What was most likeable about Walt Disney was that he was relatable; he came to embody the American values of courage, determination, wholesomeness, innocence, imagination, and self-confidence. Although Walt Disney was influenced by the setting in which he grew up, he shaped the American pop culture and everyday life of virtually the entire twentieth century, and he supported our country in times of need; his legacy continues today. Walt Disney was born on December 5, 1901 in Chicago, Illinois, but his family moved soon after his birth. Although Walt Disney shaped most of twentieth century pop culture, he himself was influenced by his small midwestern hometown: Marceline, Missouri. Walt Disney only lived there for a few years as a child, but it still had a major impact on his life and career. It was a traditional, Midwestern town which made Disney just an average guy, relatable to any other typical American. This ordinary town lacked magical and fantastical features, which could have contributed to his craving for magic and fantasy later in life. He grew up around animals on a farm; so many of his early animations consisted of animals and rural themes. He used a lot of â€Å"outhouse† and farm humor in his cartoons such as outhouse gags, goosing gags, bedpans, Johnny-pots, thinly disguised farts, and cow udders. Even though Disney grew up in a traditional American small-town, his childhood occurred during the onset of the decline of small-town America. Disney loved small-town America so much that he wanted to preserve and live by its values, such as self-determination and hard work, because they were fleeting so fast. Disney brought these values to everything he did in life and tried to encourage them in others. Walt Disney’s first claim to fame was the animated character, Mickey Mouse. Disney created Mickey Mouse in 1928, and from the very moment Mickey hit the public eye he appealed to all Americans. Mickey’s story of â€Å"rags to riches† touched the hearts of many Americans and gave them inspiration to follow their dreams. He was memorable and loveable in that he was the stereotypical hero, coming out victorious in many difficult situations. His courage, strength, will, ingenuity, and faith in himself allowed ordinary Americans to relate to him, and brought hope to them in troubling times. Mickey played many different roles, so many different people could relate to him. Mickey was know all over the world, – Michael Maus in Germany, Michel Souris in France, Miki Kuchi in Japan, Mikkel Mus in Denmark, and Miguel Ratunocito in Spain- and eventually became one of the most well known symbols in the entire world. Mickey Mouse helped bring the world a little closer together, and brought joy and courage to people in times of need. The Great Depression was one of the bleakest periods in American history, but it became a bit more enjoyable with a little help from Walt Disney’s Mickey Mouse Club. The original purpose of this children’s club was to attract more young moviegoers with discounted ticket prices, and its first theater-based club meeting was on January 11, 1930 at the Fox Dome Theater in Ocean Park, California. It gave kids something fun to do during the depression. â€Å"By 1932, there were already over a million boys and girls belonging to the Mickey Mouse clubs all over America. † Local businesses benefitted from the club too: bakeries, ice cream shops, drug stores, and banks all donated prizes for the kids. The free goodies won the kids’ hearts, and the local businesses gained a loyal customer base. The Mickey Mouse Club made kids feel like they were a part of something; at the start of every meeting the club members would recite an oath, and each club member received an ID card with the oath printed on it upon joining the club. Like the Boy Scouts, the Mickey Mouse Club taught kids how to be good and useful citizens, truthful, honorable, to follow their dreams, respect their elders, and take care of the aged, helpless, and smaller children. The Mickey Mouse club instilled values in the children of the depression, which were then carried through the rest of the century and helped make good citizens out of generations of children. Walt Disney’s empire grew from a small â€Å"studio† over a garage to a world famous corporation. Walt Disney Studios was built in 1939 in Burbank California, specifically for the animation process. As Disney Studios became more popular, they began selling products featuring Mickey Mouse, Donald Duck, and other familiar Disney cartoon characters. All of America was covered with Mickey Mouse wallpaper, dishware, cookie jars, ashtrays, salt and peppershakers, watches, clothes, wallets, pocketbooks, lunchboxes, and other household items. Mickey Mouse infiltrated every home and became a part of everyday life. Disney merchandise helped stimulate America’s economy during the Great Depression; people may have been short on cash, but they always had enough for Disney merchandise. Disney sold approximately twenty million dollars worth of merchandise from mid-1933 to mid-1934. Disney even saved several companies from going bankrupt during the depression. Ingerscroll-Waterbury got the license to sell Mickey Mouse watches, and Lionel Corporation got the license to sell Mickey and Minnie themed electric train toys. These companies were saved from bankruptcy because people could not resist buying anything Mickey Mouse-themed. In the 1950’s the Mousecap was the primary symbol of Mickey Mouse pop culture. Children were proud to wear them and say that they were official members of the club. That decade Mickey â€Å"appeared on five thousand different items, which had contributed a quarter of a billion dollars to the gross national product. Mickey Mouse became an authoritative figure in the merchandise world. Whatever companies Mickey Mouse endorsed, such as General Food, Standard Oil, National Biscuit Company, and National Dairy Products, consumers would always buy their products. Numerous of companies fought to get Mickey Mouse as their spokesperson. Liquor, cigarette, and pharmaceutical companies vied for Mickey’s support, but Disney would not sponsor anything that went against Mickey’s wholesome image. Mickey Mouse had a huge influence on the merchandise industry of the twentieth century, and in doing so Disney Studios extended its power to the economic and everyday part of American life. During World War II Walt Disney expanded his studios to play a role in American politics. After the attack on Pearl Harbor the U. S. government commissioned Disney Studios to create military training videos and propaganda films. Since Mickey Mouse was so popular the government thought they could make military training a little more enjoyable by employing Mickey Mouse and other Disney characters for the movies. Disney Studios became the first and only Hollywood studio the army took over, when 500 US army troops were stationed there. They stored repaired equipment in the soundstage and held ammunition in the sheds. President Franklin Roosevelt used Mickey Mouse as an international symbol of peace to improve the U. S. ’s Good Neighbor Policy; Walt Disney led a Goodwill Tour in South America in 1941 where a vast amount of kids who did not even speak English were eager to meet the creator of Mickey Mouse. Disney was also commissioned by the United States Treasury Department to create a film that would encourage people to pay their taxes. Mickey Mouse appeared as an Uncle Sam type figure and was the symbol of American patriotism during World War II. Mickey was the star of many U. S. nd Allied war posters and propaganda. He appeared on at least 35 home-front insignia designs created by Disney Studios, and he was the mascot for the Red Cross during the war. Disney and Mickey Mouse were such a big part of American culture and the war effort that â€Å"the password for the Allies who stormed the beach at Normandy to crush the German army was ‘Mickey Mouse. ’† Even people on t he Axis Powers looked to Mickey for inspiration during the war. Walt Disney’s help in the war effort further incorporated Disney into the pop culture and everyday life of Americans. One of Walt Disney’s greatest contributions to American pop culture was the amusement park, Disneyland, in Anaheim, California. With Disneyland Walt Disney changed the whole idea of an amusement park. It was not simply a park; it was a whole world; a full imaginative experience. Building Disneyland allowed Walt Disney to live out his imagination, and visiting Disneyland allowed every person to live out his or her wildest dreams. Disneyland appealed to everybody. The park’s traditional turn-of-the-century American main street- Mainstreet USA- made everyone feel at home. Disney filled the park with iconic American images along with images that he had created such as Cinderella, Snow White, the Three Little Pigs, and Mickey and Minnie Mouse. Disneyland exemplified how much Walt Disney had influenced the imaginations of Americans by creating a certain psychological experience in the park; it was not too loud, crowded, or chaotic like other parks, and everything was soft, harmonious, and unthreatening. The park was also appealing in its cleanliness, the efficiency of the lines, the weather, and even the sound of the park. Disneyland manipulated people into being happy by bringing out the child in everyone, but people were fine with the fact that they were being manipulated because it was executed so well. Disneyland also provided a lesson in American heritage; it taught Americans to remember their roots and traditional post-war values before they go on to fulfill their dreams. Disneyland was also a reflection of Walt Disney himself. Walt Disney put every single one of his living fibers into his park. He micromanaged everything and obsessed over every little detail until it was perfect. His commitment to the quality of the park was one thing that made it so successful; he continuously improved the park, came up with new ideas, new angles, and new additions to make Disneyland more attractive. He felt like this park was another chance to create his fantasy world and make everything exactly how he wanted it. Disneyland portrayed Disney’s sense of wish fulfillment, and life experiences and journey with the use of the different worlds of Disneyland. The images and different lands created in Disneyland could also be applied to the history and growth of America. Frontierland displayed traditional turn-of-the-century images and transported the guests back in time. Fantasyland allowed Americans to live out their wildest dreams and imagination. Adventureland depicted America’s desire for excitement and new experiences. Tomorrowland illustrated the promise of coming technological advances and America’s desire for progress. Disneyland was more than just an amusement park: it affected the lives of generations of people. Walt Disney had such a great effect on people because he embodied the idea of a true American. He represented the typical self-made, family man that people could look to and think if he can make it big, then so can I. Disney was simple, clean, moral, and innocent, and these qualities were infused into his animations and transmitted to people. His old-fashion values such as hard work, perseverance, generosity, and integrity emanated from everything he did and people were just drawn to him. These qualities and values gave people what they needed in troubling times: hope and the ability to believe in themselves. Disney’s personality made him a beloved figure in millions of middle-class households, and he affected the lives of many Americans. Walt Disney died on December 16, 1966, but his legacy continued long after he was gone. In Disney’s mind his greatest legacy would be his work with the Chouinard Art Institute in Los Angeles, California. Mrs. Nelbert Chouinard created the Chouinard Art Institute in 1921 with the belief that an art school was needed on the west coast of the United States. Disney had been interested in the Chouinard Institute since the 1930’s and when Mrs. Chouinard fell ill could not take care of it anymore he took over the institute. Disney knew how difficult it was to get admitted to art school and become successful, so he wanted to help out aspiring artists as much as he could. He financed the school, expanded it, and eventually combined it with the Los Angeles Conservatory of Music and renamed it the California Institute of the Arts. Walt even said that CalArts would be what he is remembered for. Walt Disney’s legacies continue to have an ongoing impact. Disney’s brother, Roy, once said about Walt Disney, â€Å"There is no way to replace Walt Disney. He was an extraordinary man. Perhaps there will never be another like him. † He distracted people from the strife of the Great Depression, helped our country during World War II, and provided support afterwards. He taught children how to be responsible citizens, while still allowing them to have fun and follow their dreams. He reinforced traditional American values into the minds of Americans. He helped connect the cultures of the world through the use of an iconic cartoon image, while making American culture the most dominant. He recreated the idea of an amusement park and encouraged wish fulfillment. Most importantly he demonstrated how one could turn dreams into reality. Walt Disney inspired the live of millions of children and adults all over the world, and he is considered one of the most influential people of the twentieth century.
